You would need to connect the PC USB to a Pi OTG USB port to make that work. You cannot plug Pi USB-A into PC USB-A. It won't work and it may let the magic smoke out of one or both.Right now if I press an key on keyboard or click on mouse then it wake-up, so it is working.
So I think to connect an male-male USB 2.0 type A cable to the RPi USB port and from there to send the "magic" signal in order the PC to "think" that mouse/key were pressed and to wake-up itself.
A better idea IMO is to use a Pico W to act as USB HID keyboard. When it receives a command over Wi-Fi, Bluetooth, even UART, it can send a key press to the PC and wake it up.
